Determining myocardial energetics (high energy phosphates contents, ATP, PCr synthesis, substrate selection simultaneously with heart functions at real time and intracellular pH, Na at whole heart level).

Overview

Our NMR system is a powerful tool to determine myocardial energetics (high energy phosphates contents, ATP, PCr synthesis, substrate selection simultaneously with heart functions at real time and intracellular pH, Na at whole heart level).

We can also simultaneously assess glycolysis and pentose phosphate pathway at a single sample At the end of the NMR experiments, the hearts can be freeze-clamped for other molecular measurement and HPLC study.

Physiological NMR Core is equipped with 400MHz NMR, Langendorff system and Water HPLC system, and currently provides the following services:

1. Real-time and simultaneous determination of cardiac energetics, intracellular pH, Glucose uptake rate, ATP and PCr synthesis rates with function in perfused organs with 31P-NMR
2. Simultaneous determination of glycolysis and pentose phosphate pathway with 13C-NMR
3. Track substrate utilization/metabolism with 13C-NMR
4. Determine of intracellular Na content with 23Na-NMR
5. Simultaneous determination of cellular redox potentials, energy metabolites and nucleotide catabolites in a single sample with HPLC
6. Enzymatic determination of L2HG and D2HG
